The nine-time Tony award winner replaces David Cassidy as star of the special effects show on Jan. 15. Cassidy decided to leave the show after two years.
Tune, 59, will play himself in the show, which has been changed to showcase Tune's tap dancing and singing.
"Everyone dreams," Tune said. "And I've lived my life in constant pursuit of my dreams, including the one to headline in a big Las Vegas spectacle."
Tune made a name for himself in Broadway shows such as "The Best Little Whorehouse in Texas," "Seesaw," "Nine," "My One and Only," "Grand Hotel" and "The Will Rogers Follies."
